No, unfortunately you have not qualified.  Novice wins are conformation wins but are not a qualifying win for HOY.  The only time a Novice win becomes a qualifying win is if you proceed to the Championship class and win this or win Reserve Championship.  Please note that you will need 2x Reserve Championship wins in order to qualify.

I have one win in Mare and One Reserve Championship in In Hand - does this mean I qualify for HOY?
No, unfortunately you have not qualified.  You will need another Reserve Championship OR another Conformation win in order to qualify.
